18-month post-doc position with an expected starting on October 1st,2024.
As a Servier collaborator, the candidate will be part of the BioTransformation & BioDistribution team in the DMPK department and will work in our whole new the R&D Paris-Saclay Servier Institute (Gif-sur-Yvette). He/she will be a key actor of the Mass Spectrometry Imaging platform and will have access to cutting-edge equipment’s in the biodistribution lab (Solarix 7T (Bruker), DESI-Xevo G2 XS (Waters), M5 Sprayer (HTX), microtomes (Leica)).
This project will be divided in 3 main work packages:
WP1: Atlas of detectable endogenous metabolites.
The aim of the WP is to build an atlas of detectable endogenous metabolites in various tissues using the different instruments and methods available at Servier.
The candidate will have to select the relevant samples for Servier application, i.e. which organ (brain, tumor, pancreas…), which conservation process (Fresh Frozen, FFPE…) and will analyze them with the different methods available (DESI-ToF +/-, MALDI-FTICR +/- with various matrix). He will then have to analyze the data to identify the list of endogenous metabolites detectable in each condition. He/she will then elaborate a clear decision tree for technology selection based on the type of tissue and the biomarkers of interest to solve biodistribution questions on our therapeutic projects.
WP2: Implementation of new tools for biodistribution studies by MSI
To accelerate our project and strengthen our catalogue in MSI, the candidate will implement new technologies and methods to our platform, starting with the Myralys® technology from Ambergen. This will enable protein and oligonucleotide detection by MALDI using the same slide as the one used for the classical MALDI-MS acquisition. The candidate will also be responsible for technology watch to explore new possibilities.
WP3: Imaging of brain/tumor organoids coculture in collaboration with Institut Goustave Roussy.
To image the metabolic and lipidic change occurring during neural cell differentiation and tumor invasion, a collaboration with IGR is currently being set up to perform some MALDI experiment (method development and image acquisition) on brain organoids.
During his/her post doc, the candidate will also be one of the referent people for the Mass Spectrometry Imaging activities. He/she will bring his technical expertise to support other researchers willing to use the MSI platform for their own project around biodistribution and multimodal imaging. He/she will work in close collaboration with other internal department around imaging multimodalities (including histopathology, spatial transcriptomics and proteomics) as well as with external partners.
Responsabilities :
- Build an atlas of endogenous metabolites to elaborate a decision tree for the selection of the right MSI technology in upcoming projects
- Implement new technology in our internal platform such as Myralys® technology
- Technical responsibilities of lab instrument in interaction with team’s scientists and instrument manufacturer engineer
- Ensure good communication with other partners to support projects involving MSI
- Participate to the BioTransformation and BioDistribution team activities
Qualification :
PhD in analytical chemistry or related discipline
Hard skills :
- Mass Spectrometry Imaging (mandatory)
- High Resolution Mass Spectrometry of small molecules (Spatial) Metabolomics
- Data processing (SCiLS)
- Skills in programing and large molecule analysis (e.g.: Antibody Drug Conjugates, Oligonucleotides...) would be a plus
